چکیده

Importance and challenges of identifying body fluids in forensic casework In forensic casework, correctly identifying body fluids such as blood, semen, or saliva is important for reconstructing a crime scene. Analysis of stains or biological fluids recovered during an investigation may provide potential sources of DNA that can be used to identify the donor of the fluid or stain [1]. However, body fluid samples can be difficult to work with, as often only limited quantities of sample are available at crime scenes. Moreover, samples may have been exposed to conditions that contribute to the degradation of RNA, and may yield only minute amounts of low-quality genetic material. Traditional serological or immunological methods of identifying body fluids can be costly in terms of time and labor, and require large amounts of sample. Moreover, these protein-based assays have varying degrees of sensitivity and specificity [2]. To be suitable for forensic casework, a body fluid identification assay must:

Characterising the fluctuation of microRNA expression throughout a full menstrual cycle

When processing a crime scene, obtaining a DNA profile that can identify an individual is extremely important. However, the identification of the body fluid that the sample was obtained from could provide key information of the events that occurred. microRNA (miRNA) expression analysis is a technique that has the potential to differentiate body fluids. The presence and expression of body fluid ...

Real-time quantification of microRNAs by stem–loop RT–PCR

A novel microRNA (miRNA) quantification method has been developed using stem-loop RT followed by TaqMan PCR analysis. Stem-loop RT primers are better than conventional ones in terms of RT efficiency and specificity. TaqMan miRNA assays are specific for mature miRNAs and discriminate among related miRNAs that differ by as little as one nucleotide. Furthermore, they are not affected by genomic DN...

Evaluation of stem-loop reverse transcription and poly-A tail extension in microRNA analysis of body fluids.

MicroRNA has been demonstrated to be a viable tool for body fluid identification purposes in forensic casework. Stem-loop reverse transcription (slRT) is regularly used for cDNA synthesis from mature miRNA, along with poly-A tail extension. Both have been used in a forensic context, but no direct comparison has been carried out. It has also not been shown whether poly-A tail extension can be us...

A cerebrospinal fluid microRNA signature as biomarker for glioblastoma

PURPOSE To develop a cerebrospinal fluid (CSF) miRNA diagnostic biomarker for glioblastoma. EXPERIMENTAL DESIGN Glioblastoma tissue and matched CSF from the same patient (obtained prior to tumor manipulation) were profiled by TaqMan OpenArray® Human MicroRNA Panel. CSF miRNA profiles from glioblastoma patients and controls were created from three discovery cohorts and confirmed in two validat...

The Potential Use of Tissue-Specific MicroRNAs for Forensic Sample Identification,

Background: In forensic casework, the identification of human material, especially at the scene can be extremely important. Several methods exist to date, but sometimes they lack sensitivity and specificity at detecting biological stains from human sources. Forensic RNA research using mRNA markers is still in its infancy but has provided the possibility of human stain identification. However, t...
